  • Cell culture is the process of growing cells outside the body ‘in vitro’ as opposed to inside the body ‘in vivo’. Cells could be immortal cells such as cancers see HeLa or samples of tissue taken surgically or primary cell lines (cells that have not been cultured very much before). Cell culture could be liquid or semi solid or solid. The medium is usually a broth of nutrients, growth factors and buffers (salt which counteract the acidity produced by cell waste products). The medium also has a colour acidity indicator to help the acidity of the medium. Red neutral yellow acidic. Cells are incubated in a err incubator that is humid and has co2 gas added. This mimics the in vivo situation where CO2 is raised compared to atmospheric. The manipulation of cells in culture must be carried in a laminar flow hood this is basically a bench that has filtered air passed over it. This keeps microbes out of the hood and out of the lovely rich media which would grow bacteria if given half the chance.everything must be sterile or filtered to prevent contamination by bacteria which would out grow and kill any cells. Growth media will also contain antibiotics to help this. Final word on growth medium it usually contains a supplrment of fetal calf serum which was found to have all the necessary growth factors for mammalisn cells.
  • Collect sample of cells, place in growth medium with agar and liquid growth medium, grow by mitosis
